Leeds will be hoping to avoid another embarrassing FA Cup exit when they travel to South Wales (12pm).
The Whites were dumped out of the competition in Round Four last year by non-league Sutton United.
Leeds, who currently occupy the final Championship play-off spot, and Newport, 11th in League Two, are separated by 53 places in the English football pyramid.
The two sides faced each other in the Carabao Cup in August with Leeds running out 5-1 winners.
Whites head coach Thomas Christiansen is expected to rotate his side for the trip to Rodney Parade, with a number of youngsters set to feature.
"We have a difficult game with long travel," he said.
"A team on their pitch, it will be direct play and a tough game away for us to play but we need to go into the next round, this is obvious and we will do our best."
Newport boss Michael Flynn said: “Leeds are a huge club and we are lucky and privileged to have them at Rodney Parade.
“We’ve drawn them in two successive cups and now finally get them at Rodney Parade.”
