The England international secured a 2-2 draw after Nathan Dyer was harshly judged to have hand-balled in the area just as Claudio Ranieri's side look set to pull off a shock win at White Hart Lane.
Kane's landmark came in only his 104th game, but Pochettino insisted there was nothing special about taking a penalty.
"Good for him," he said. "I'm happy. The goal he scored helped us to stay alive in the FA Cup.
"But there are a lot of players who can take the responsibility of a penalty. It's not a physical quality. Obviously, though, if Harry Kane is on the pitch he's the first to take it."
