PSV are undefeated this season and beat Eredivisie minnows Cambur 6-0 at the weekend, while United toppled rivals Liverpool 3-1 at Old Trafford.
And De Jong, who boasts five goals for the club already this term, insists focus is the key to success for the hosts.
“I am raring to go,” said the captain. “This is a beautiful podium to show the world what you can do.
"It is a nice game to play and we are facing an amazing opponent. It would be fantastic if we could spring a surprise.
“They have a number of players with a lot of individual quality. We cannot afford to lose focus.
"They make the most of each minor mistake. We have to remain alert for the full 90 minutes."
