The world number one, who turned 30 yesterday, lost in straight sets - 6-2 6-4 to Fabio Fognini.
The Rome Masters was one of nine titles Murray won last year, but he was dumped out this time in the second round.
Fognini, who is ranked 29 was more consistent in all areas of his game.
A first serve percentage rate of 64 compared to Murray’s 48 highlighted the gulf in class.

With the French Open getting underway on May 28, Murray doesn’t have long to turn his form around.
Meanwhile, Murray’s compatriot Aljaz Bedene was also knocked out in the second round.
He was taken out by Novak Djokovic - 7-6, 6-2.
