The quarterback suffered the injury early in the second quarter when he took a big hit.
He immediately stayed on the floor and held his right leg in severe pain.
Knights and Bulls players knelt down beside Milton before he was stretchered off with what looks like a serious injury.
The star quarterback has been instrumental for the Knights, helping them to 23 straight victories over the last two seasons.
ESPN opted to not show replays of the incident but video footage soon emerged on Twitter.
So far this season, Milton had thrown for 2,577 yards and 25 touchdowns, including another nine touchdowns on the ground.
He was transported to St. Joseph Hospital after the incident.
The Knights are aiming to make the New Year's Six bowl for the second straight year.
The junior from Hawaii started in 2016.
He made big progress last season as UCF declared itself national champions following a win over Auburn in the Peach Bowl.
Milton is seen as one of the best quarterbacks in the country over the last two seasons.
