The centre-half was withdrawn at half-time in Manchester United's 4-1 victory over Fenerbahce in the Europa League.
Marcos Rojo replaced the England international, and it appears Smalling's chances of featuring at Stamford Bridge are now slim.
The 26-year-old has started Manchester United's last four Premier League games, largely due to the absence of Luke Shaw.

But Mourinho revealed after tonight's clash that Smalling was taken off as a precaution and he could miss the crunch clash in less than three days' time.
"He had a bad muscular feeling," Mourinho said on Smalling.
"We were 3-0 up so we decided to change.
"Hopefully he can recover for the weekend."

With Shaw back fit, having played the full 90 minutes at Old Trafford, Daley Blind is expected to come back in on Sunday with Smalling doubtful to be 100 per cent ready.
However, amid the injury concerns, Smalling did deliver a positive message to fans after the game.
He tweeted: "Good win tonight! All eyes on Sunday now!"
The defender gave no indication to his supposed problem, but Mourinho will want everyone sharp for the showdown with former club Chelsea.
