United were already without Ashley Young, who is sidelined with a groin injury sustained in last week's win at Liverpool.
And things went from bad to worse for Louis van Gaal when Darmian was stretchered off when making a brave headed clearance.
The Italian, who moved to United from Torino in the summer and has featured on both flanks at full-back in his debut campaign, was replaced by Paddy McNair.
Another crocked defender leaves Cameron Borthwick-Jackson as Van Gaal's only fit full-back.
Luke Shaw (broken leg), Marcos Rojo (dislocated shoulder) and Antonio Valencia (ankle ligaments) are the other senior full-backs on the sidelines.
United are also without Phil Jones and Bastian Schweinsteiger.
