The City boss' comments came after the Reds salvaged a dramatic three points against Aston Villa last weekend, with the Senegal star going to ground easily in the first half as the visitors chased the game.
Mane was shown a yellow card for simulation after hitting the deck in the 37th minute under pressure from Frederic Guilbert, with replays showing there was little contact between the pair.
After the incident, Man City boss Guardiola said of Mane: "Sometimes he’s diving, sometimes he has this talent to score incredible goals. He’s a special talent."
And those remarks did not sit well with former Borussia Dortmund boss Klopp, who snapped back in the Reds' pre-Champions League press conference on Monday.
Ahead of the visit of Genk, the boss said: "I'm not really in a Man City mood at the moment.
"Do I like that he says this about one of my players? I'm not even 100 per cent sure if he spoke about Sadio [Mane] or us in general. I didn't hear Sadio's name.
"I don't know how he could have known about any incident in the game so quickly after [Man City's win over Southampton].

"Sadio is not a diver. There was a situation in the Aston Villa game where he got a contact and went down. Maybe it wasn't a penalty but there was contact - it's not like jumping over a leg and acting like he hit you."
And City fans believe Guardiola has managed to get a rise out of his opposite number, handing the champions an upper hand in the title race.
One supporter tweeted: "We’ve absolutely rattled him."
Another added: "LOOOOOL Pep got mans on strings out here."
