Klitschko lost to Tyson Fury last year, breaking an 11-year unbeaten streak, and has not fought since.
However, Fury has since vacated the WBA and WBO heavyweight titles after admitting to cocaine use while suffering depression.
The WBA has agreed to sanction the fight, which was originally slated for December 10 in Manchester before the Ukrainian pulled out with a minor injury.
But now Matchroom promoter Eddie Hearn has confirmed that the fight will indeed take place on April 29 next year.

The bout will take place at Wembley Stadium and could break a record for attendance at a boxing match.
"There is no doubt this fight must happen," Klitschko said.
