Leader of Labour Leave Graham Stringer said it is an “attack on democracy” for former Labour MP Sadiq Khan to call for a second Brexit referendum.
Speaking to LBC on Sunday, Mr Stringer said: “I think it’s a complete and straightforward attack on democracy.
“Everybody who voted in the referendum had every expectation on whatever the result it would be carried out.
“Not just the great debate at Wembley but at the second bill the put a referendum into law, not one person said that if the negotiations with the EU get difficult then we will have another referendum.
I think it’s a complete and straightforward attack on democracy.
“In fact, Hilary Benn, Labour’s spokesperson, said this is up to the people to decide.
“What is happening now is a significant number of people who were Remainers, Sadiq has always been a Remainer, are trying to undermine the democratic vote.”
Labour mayor of London Sadiq Khan hit the headlines on Sunday after backing a second Brexit referendum, which he suggested would rule out a hard Brexit option.
Writing in The Observer, Sadiq Khan claimed evidence signalling Brexit would "result in fewer jobs, less prosperity and a reduced role for Britain on the world stage" had become "irrefutable," urging the Prime Minister to organise a second referendum.
Mr Khan, a former Labour MP for Tooting and staunch Remainer, argued there were only two possible outcomes in the Brexit talks: a bad deal or “no deal” at all, which he claimed would be even worse.
He said: “Both these scenarios are a million miles from what was promised during the referendum campaign, only further exposing the lies and mistruths sold to the public.
“They are both incredibly risky and I don’t believe Theresa May has the mandate to gamble so flagrantly with the British economy and people’s livelihoods.”
The latest push from one of Labour's most powerful figures will likely increase the pressure on Jeremy Corbyn ahead of the party conference next week.
There are already more than 100 anti-Brexit motions submitted, including calls for a new vote on the final deal.

Among the Brexiteer wing of the Tory party, there is a growing movement urging the Prime Minister to "chuck Chequers".
Former Brexit minister Steve Baker has warned Mrs May that up to 80 Tory MPs are prepared to vote against Mrs May's Chequers plan.
Despite criticism from within Mrs May's own party, No 10 confirmed its plan was the "only one on the table" able to deliver on the will of British people while avoiding a hard border.
The Chequers agreement in July triggered the cabinet resignations of both David Davis and Boris Johnson.
