The Hollywood star has been given odds of 4/9 of triumphing on the night, after being nominated for his role in The Revenant - also up for Best Film.
If bookmakers William Hill are correct in their predictions, the 41-year-old will beat Brit Eddie Redmayne, who won Best Actor at last year's event.
The Revenant, which also stars Tom Hardy and Will Poulter, follows Leo's legendary frontiersman Hugh Glass on his quest for survival during a fur trading expedition in the 1820s.
The five-time Oscar nominated actor also faces stiff competition from Michael Fassbender (Steve Jobs), Bryan Cranston (Trumbo) and Matt Damon (The Martian).
