Europe came into the final day at Le Golf National with a 10-6 lead following a stirring comeback after Friday morning's disappointing fourballs session.
And Molinari had played an integral role in securing that advantage, striking up an incredible partnership with Ryder Cup rookie Tommy Fleetwood.
Molinari was in singles action on Sunday in something of a mismatch with USA veteran Mickelson.
And in the end the Italian watched on as Mickelson's blunder handed Europe the point they needed to regain the Ryder Cup trophy.
After Molinari had found the green on the par 3 16th hole, Mickelson stepped up to take his own tee shot.
But the five-time Major winner misjudged and looked resigned to defeat as his ball flew towards the water.
Mickelson's error sparked jubilant scenes from Molinari and the watching Europe supporters.
Molinari's 4&2 match win capped off a superb weekend of golf for the 2018 Open winner.
Ecstatic Europe captain Thomas Bjorn hailed every member of his 12-man squad following the victory at Le Golf National.
"I’m probably a bit prouder today than I was on Thursday evening," Bjorn said.
"It's been easy - they’ve been amazing; all 12 of them have been unbelievable.
"They were so determined. They set out to do a job, so it was easy for me to guide them in that direction. They stood up to it.
"So many things have happened this week, so many great stories but it's down to 12 players and them only.
"This is such special event, when you come in with right attitude it's such a great thing. I couldn’t believe they would stand up like they did, but they did because of the 12.
"[I should] wrap them up and take them to America for the next one. It was getting tough but in general I had belief and faith in the team that they were going to deliver.
"It's the Ryder Cup. They come away from home and it’s tough. Our team wanted it so much this week - that’s why they did what they did."
