Swansea boss Guidolin will become the latest incumbent in the Vicarage Road hot seat since Gianfranco Zola resigned in December 2014.
The Hornets current boss, Quique Flores, 51, will be axed when owner Gino Pozzo activates a break clause in his contract.
Despite taking Watford to the FA Cup semi-final and ensuring their place in the Premier League for next season, it won't be enough to save Flores' job.
Meanwhile, Brendan Rodgers will replace Guidolin, 60, at the Liberty Stadium.
