Wanyama, 24, reportedly told the club last week that he wants to leave before the transfer window shuts.
Premier League rivals Arsenal and Tottenham are believed to be keen on signing Wanyama, who Saints value at £18million.
The combative midfielder looked to be on his way out after failing to make the squad for tomorrow night's trip to FC Midtjylland, but reports claim he is missing with a stomach infection.
And, according to ESPN FC, the St Mary's outfit have now informed Wanyama that he will not be allowed to leave.
The Kenya international is understood to want a move to White Hart Lane to work under his former boss Mauricio Pochettino.
Southampton have already been forced to sell key players Nathaniel Clyne and Morgan Schneiderlin to Liverpool and Manchester United, respectively, this summer.
