Holt bundled home Emmerson Boyce’s effort for Wigan’s second goal.
Ben Watson got the ball rolling with a 25-yard effort, his first goal since winning the FA Cup for Wigan at Wembley.
But Holt was the focal point in everything Wigan did as they looked a class above the 10-man Tykes, who saw sub Dale Jennings’ debut last only five minutes before he was given a straight red card for a lunge on James McArthur.
With 10 minutes left Leon Barnett fired home from close range, then Shaun Maloney weighed in with a strike as the scoreline finally started to reflect Wigan’s dominance.
There would have been few complaints had Wigan bagged a few more as Barnsley were steamrollered by Owen Coyle’s lively new-look team.
