The pop star will perform classic tracks and hits from Joanna at the 51st Super Bowl at the NRG Stadium in Houston, Texas, on February 5 2017.
It comes after the singer said that giving a rendition of the US national anthem last year was a highlight of her career.
“This is one of the highest honours of my career,” she explained to the NFL Network at the time.
She added: "I get a chance to sing for all the athletes - who have been working so hard their whole lives for this moment - the coaches, as well as the fans in the stands who are waiting for this moment.
“I think it marks what being an American is all about."
Lady Gaga follows last year’s half-time headliners Coldplay, who were joined by Beyonce and Bruno Mars on stage.
Following the performances only three people turned up for an anti-Beyonce protest in New York.

Back in August Adele admitted she was offered the spot, but turned it down.
She said: “That show is not about music. I can't dance or anything like that. They were very kind, they did ask me, but I said no.”
Lady Gaga also is rumoured to be headlining Glastonbury next year, contending with The Stone Roses, Daft Punk, Guns N’ Roses, Ed Sheeran and Radiohead.
